The dynamics in the diverse microflora directly associated with humans, and associated effects on human health, are just beginning to be understood , and the impacts of isolation and space conditions (particularly ionizing radiation) are poorly understood. Just as on Earth, microorganisms will play a key role in sustainably providing environmental life support within the system, including water purification, nutrient regeneration for plant systems, and potential energy and material production. Innovative, small scale biological systems are unique sustainable attributes, and their application in spaceflight offer a model for decentralized systems on Earth. While various conceptual and empirical approaches for incorporating microorganisms into life support systems for space have been evaluated (and will be reviewed) , the long term stability of integrated systems have not been established. The direct dependence of human health on the complex microbiome within the closed systems challenges our ability to construct and maintain microbial communities. Advanced monitoring tools may be useful, but conceptual frameworks of microbial communities as complex adaptive systems are even more important for effective management.

The Physical Virology GRC covers viruses as paradigms for nanoscale assembly, transformation, and the properties of protein cages on that size scale as well as their applications. The principles underlying virus-inspired systems organization have been investigated by mathematicians, physicists, chemists, material scientists, and biologists. This diversity is a strength because of the breadth of perspectives but poses challenges because of the poor communication between disciplines. Biological efforts to prevent HIV infection center on four independent approaches: inducing adaptive immune responses through vaccination, augmenting innate responses, using peri-exposure prophylactic drug therapy, and developing microbicides and/or recombinant antiviral microbial products. While all four of these approaches have promise, they still require significant optimization and further clinical trials. Indeed, successful prevention of HIV infection will likely require a combination of these approaches. Such development efforts require a better understanding of viral and immunological events at the site of transmission. This Keystone Symposia meeting will have a focus on mucosal immunology as well as the interplay between the virus and innate and adaptive immune responses, particularly during the acute phase of the infection. Leading experts in the fields will discuss recent scientific advances in these varied approaches to preventing infection and present data from recent clinical trials testing their efficacy.

The mechanisms underlying the pathogenesis of influenza remain controversial. The direct cytopathic effects of viral replication, tissue tropism of the virus, viral-bacterial synergy, as well as innate host responses are inextricably linked and play roles to varying degrees in "seasonal," zoonotic and pandemic influenza, examples being the pandemics of 1918 and 2009 and H5N1 avian influenza. Animal models, though indispensible, have significant limitations with regard to physiological relevance to human disease. The current symposium brings together researchers working on the virus, viral receptors and tissue tropism, innate and adaptive immunity, systems biology and clinical aspects of lung injury and host defense, to address questions on the pathogenesis of influenza. The aim will be to integrate data from animal and ex vivo / in vitro human experimental models as well as human disease to understand pathogenesis of influenza and how this may lead to effective interventions. As this symposium will take place in the aftermath of the first pandemic in 40 years, there will be a wealth of new knowledge as well as intense scientific interest in the subject. In view of the particular interest in influenza and other viral respiratory diseases generated in the Asia-Pacific region arising from the avian flu H5N1 and SARS experience, situating the meeting in Hong Kong would be particularly appropriate.

Cryptococcus neoformans is an environmental microorganism that has become a major threat to a growing number of immunocompromised patients in today's world. Recently, the CDC estimated more than 1 million new cases/year of cryptococcosis will occur worldwide in patients with AIDS, with more than half of those affected by cryptococcosis dying of the infection.Importantly, during the last few years, cryptococcosis has emerged in the Vancouver Island, Canada and Pacific Northwest of the US as an important disease also in immunocompetent subjects. These epidemiological data underscore the potential for this microorganism to continue to emerge in unexpected geographic and clinical settings.
